چکیده

The authors report on fast phase imaging in liquids achieved by fast phase detection in rapid scan atomic force microscopy. The phase-shift images clearly revealed the compositional heterogeneities in styrene-butadiene-styrene block copolymer films even at an imaging rate of less than 100 ms/ frame. They found that the contrast variations of the phase images depended on the phase-shift detection timing within a single oscillation cycle. The phase contrast increased as the tip approached the surface in each oscillation during imaging, while it decreased as the tip withdrew from the surface. © 2006 American Institute of Physics. Origins of phase contrast in the atomic force microscope in liquids.

We study the physical origins of phase contrast in dynamic atomic force microscopy (dAFM) in liquids where low-stiffness microcantilever probes are often used for nanoscale imaging of soft biological samples with gentle forces. Under these conditions, we show that the phase contrast derives primarily from a unique energy flow channel that opens up in liquids due to the momentary excitation of h...

Compositional contrast of biological materials in liquids using the momentary excitation of higher eigenmodes in dynamic atomic force microscopy.

Atomic Force microscope (AFM) cantilevers commonly used for imaging soft biological samples in liquids experience a momentary excitation of the higher eigenmodes at each tap. This transient response is very sensitive to the local sample elasticity under gentle imaging conditions because the higher eigenmode time period is comparable to the tip-sample contact time. By mapping the momentary excit...

Fast, high-resolution atomic force microscopy using a quartz tuning fork as actuator and sensor

We report a new method of achieving tip–sample distance regulation in an atomic force microscope ~AFM!. A piezoelectric quartz tuning fork serves as both actuator and sensor of tip–sample interactions, allowing tip–sample distance regulation without the use of a diode laser or dither piezo. Such a tuning fork has a high spring constant so a dither amplitude of only 0.1 nm may be used to perform...

Quantitative Force Measurements In Liquid Using Frequency Modulation Atomic Force Microscopy

The measurement of short-range forces with the atomic force microscope (AFM) typically requires implementation of dynamic techniques to maintain sensitivity and stability. While frequency modulation atomic force microscopy (FM-AFM) is used widely for high-resolution imaging and quantitative force measurements in vacuum, quantitative force measurements using FM-AFM in liquids have proven elusive...

Phase imaging of moving DNA molecules and DNA molecules replicated in the atomic force microscope.

Phase imaging with a tapping mode atomic force microscope (AFM) has many advantages for imaging moving DNA and DNA-enzyme complexes in aqueous buffers at molecular resolution. In phase images molecules can be resolved at higher scan rates and lower forces than in height images from the AFM. Higher scan rates make it possible to image faster processes. At lower forces the molecules are imaged mo...
